iPSC-Derived Glioblastoma Cells Have Enhanced Stemness Wnt/β-Catenin Activity Which Is Negatively Regulated by Wnt Antagonist sFRP4

Growing evidence indicates that cancer stem cells (CSCs) endow the tumor with stem-like properties. Recently, induced pluripotent (iPSCs) have gained increased attention because of their easy derivation and availability potential to differentiate into any cell type. A CSC model derived from iPSCs human origin would help understand driving force initiation early progression. We report efficient generation feeder-free SSEA4, TRA-1-60 TRA-1-81 positive amniotic membrane-derived mesenchymal (AMMSCs), which successfully differentiated three germ layers. then developed iPSC-derived glioblastoma multiforme (GBM) using conditioned media (CM) U87MG line CSCs U87MG, confer GBM GSC-like phenotypes within five days. Both types overexpress MGMT GLI2, but only GSCs CD133, CD44, ABCG2 ABCC2. also observed overexpression LEF1 β-catenin in both types. Down-regulation Wnt antagonist secreted frizzled-related protein 4 (sFRP4) GSCs, indicating activation Wnt/β-catenin pathway, could be involved conversion CSCs. From future perspectives, our study will creation a rapid cell-based platform for understanding complexity GBM.
